Our cells face constant stress imposed by internal and external conditions that can lead to their dysfunction, aging and death. A robust capacity to deal with such stress can prevent cellular dysfunction and delay aging, whereas susceptibility to cellular stress underlies various degenerative diseases. Our lab is interested in understanding how metazoan cells cope with stress caused by misfolded proteins in the endoplasmic reticulum (ER) and amino acid deprivation. In particular, we study signaling pathways activated by these conditions, which are referred to as the Unfolded Protein Response (UPR) and the Integrated Stress Response (ISR). We primarily use Drosophila melanogaster as a model, and this is supplemented with mechanistic studies in cultured mammalian cells. Our laboratory is one of the first to employ Drosophila to investigate UPR and ISR, and the tools that we have generated over the years are now widely employed in the field. Of particular interest is to identify the physiological roles of these stress response mechanisms in the context of neuro-degenerative disease models, in normal development, tissue homeostasis and innate immune response.
